Among two strains were exhibited laccase activity DVRSGPD-1 and DVRSGPD-2 have exhibited laccase activity and hence we have used these two cultures for further studies. Based on morphological, molecular analysis it was confirmed that the cultures DVRSGPD-1 was closely related to the sequences of Cochliobolus hawaiiensis and DVRSGPD-2 to Aspergillus niger. The sequence database was deposited in NCBI GenBank and obtained an accession number KF805051 and KX055889. Cochliobolus hawaiiensis and Aspergillus niger were screened for optimization of the medium, pH, temperature, like carbon, nitrogen, inducers, agitation, CuSo4, and concentrations for production of higher laccase activity. Cochliobolus hawaiiensis was showed the best results than Aspergillus niger in all optimization parameters, so the Cochliobolus hawaiiensis was processed for further analysis.- The laccase enzyme produced by Cochliobolus hawaiiensis isolate was purified by ammonium sulfate method and the LPVC treated with Cochliobolus hawaiiensis and resulted metabolites were subjected to SEM, FTIR, NMR, GC - MS compared with control LPVC.
